Improvement of Methods for Analysis of Mycotoxin (Trichothecenes and Fumonisins) in Beer by Liquid Chromatography Tandem Mass Spectrometry

In the previous report, we developed methods for analyzing of trichothecenes and fumonisins using liquid chromatography-electrospray ionization tandem mass spectrometry (ESI-LC/MS/MS); ("Analysis of Trichothecenes in Barley Tea and Beer by Liquid Chromatography / Tandem Mass Spectrometry"; Shokuhin Eisei Gakkai 45, 307-312 (2004); "Analysis of Fumonisin B1 and B2 in Beer and Raw Materials of Beer by Liquid Chromatography / Tandem Mass Spectrometry" Shokuhinn Eisei Gakkai 45, 255-258 (2004)). However it had a little disadvantage on the efficiency of ionization for trichothecenes, and the poor recovery rates in beer for fumonisins. In this study, we improved these methods. For trichothecenes, we introduced a multifunctional cartridge for purification and reexamined the LC/MS/MS condition. For fumonisins, we introduced a new cartridge containing mixed-mode anion-exchange and reversed-phase sorbent. As the result, the improved method for trichothecenes was more simplify and the timesaving. For fumonisins, the recovery rates spiked at the level of 5 μg/L in beer were increased (64.0%→94.6% FB1 and 52.0%→ 97.6% FB2, respectively).
